چکیده

the satellite cells. Contaminating CD45+/Mac-1+/Sca-1+ cells were subsequently removed by MACS cell separation. Cell cycle analysis revealed that over 80% of the resulting myogenic cell population was in G0, indicating a high proportion of quiescent cells in the isolated population. RT-PCR analysis showed that the expression of three Kruppel-like factors (Klf) was increased in quiescent satellite cells and decreased during a time-course of activation in culture. Furthermore, this decrease in expression correlated with the increasing expression of MyoD, which is upregulated in activated satellite cells, indicating a potential role for the Klf genes in the regulation of satellite cell quiescence.

A premeiotic function for boule in the planarian Schmidtea mediterranea.

Mutations in Deleted in Azoospermia (DAZ), a Y chromosome gene, are an important cause of human male infertility. DAZ is found exclusively in primates, limiting functional studies of this gene to its homologs: boule, required for meiotic progression of germ cells in invertebrate model systems, and Daz-like (Dazl), required for early germ cell maintenance in vertebrates. Dazl is believed to have...

Early planarian brain regeneration is independent of blastema polarity mediated by the Wnt/β-catenin pathway.

Analysis of anteroposterior (AP) axis specification in regenerating planarian flatworms has shown that Wnt/β-catenin signaling is required for posterior specification and that the FGF-like receptor molecule nou-darake (ndk) may be involved in restricting brain regeneration to anterior regions. The relationship between re-establishment of AP identity and correct morphogenesis of the brain is, ho...

A functional genomics screen in planarians reveals regulators of whole-brain regeneration

Planarians regenerate all body parts after injury, including the central nervous system (CNS). We capitalized on this distinctive trait and completed a gene expression-guided functional screen to identify factors that regulate diverse aspects of neural regeneration in Schmidtea mediterranea. Our screen revealed molecules that influence neural cell fates, support the formation of a major connect...

Cellular, ultrastructural and molecular analyses of epidermal cell development in the planarian Schmidtea mediterranea.

The epidermis is essential for animal survival, providing both a protective barrier and cellular sensor to external environments. The generally conserved embryonic origin of the epidermis, but the broad morphological and functional diversity of this organ across animals is puzzling. We define the transcriptional regulators underlying epidermal lineage differentiation in the planarian Schmidtea ...

Transcriptome Analysis Reveals Strain-Specific and Conserved Stemness Genes in Schmidtea mediterranea

The planarian Schmidtea mediterranea is a powerful model organism for studying stem cell biology due to its extraordinary regenerative ability mediated by neoblasts, a population of adult somatic stem cells. Elucidation of the S. mediterranea transcriptome and the dynamics of transcript expression will increase our understanding of the gene regulatory programs that regulate stem cell function a...
